云端論腦 第37期 預(yù)告|FGF13基因點突變引發(fā)智力障礙的機制研究 盡管許多基因變異可能與先天智力障礙 ( intellectual disability, ID)有關(guān),但導(dǎo)致先天智力障礙的基因突變在很大程度上仍不清楚。我們篩選了患有嚴(yán)重智力障礙的中國漢族兒童的基因突變,發(fā)現(xiàn)3名男性兒童在成纖維細胞生長因子13 (FGF13) mRNA 的5 '-非翻譯區(qū)(5 '-UTR)存在單核苷酸多態(tài)性(SNP)。在HEK293細胞和患者來源的誘導(dǎo)多能干細胞中,這種SNP降低了FGF13的翻譯,而FGF13穩(wěn)定發(fā)育中的神經(jīng)元中的微管。Fgf13基因5 '-UTR同源點突變小鼠表現(xiàn)為皮層發(fā)育過程中神經(jīng)元遷移延遲,學(xué)習(xí)記憶能力減弱。此外,這種SNP降低了FGF13 5 '-UTR和多嘧啶束結(jié)合蛋白2 (PTBP2)之間的相互作用,這是皮層神經(jīng)元中FGF13翻譯所必需的。因此,F(xiàn)GF13的這個5 '-UTR SNP干擾了FGF13的翻譯過程,導(dǎo)致大腦發(fā)育和認(rèn)知功能缺陷。 Title: 5'-UTR SNP of FGF13 causes translational defect and intellectual disability Link: https://elifesciences.org/articles/63021 The congenital intellectual disability (ID)-causing gene mutations remain largely unclear, although many genetic variations might relate to ID. We screened gene mutations in Chinese Han children suffering from severe ID and found a single-nucleotide polymorphism (SNP) in the 5′-untranslated region (5′-UTR) of fibroblast growth factor 13 (FGF13) mRNA (NM_001139500.1:c.-32c>G) shared by three male children. In both HEK293 cells and patient-derived induced pluripotent stem cells, this SNP reduced the translation of FGF13, which stabilizes microtubules in developing neurons. Mice carrying the homologous point mutation in 5′-UTR of Fgf13 showed delayed neuronal migration during cortical development, and weakened learning and memory. Furthermore, this SNP reduced the interaction between FGF13 5′-UTR and polypyrimidine-tract-binding protein 2 (PTBP2), which was required for FGF13 translation in cortical neurons.
